EVALUATION OF SERUM KLOTHO AND INSULIN SENSITIVITY IN OBESE PREDIABETES PATIENTS
 
Prediabetes is a state of intermediate hyperglycemia with a high risk of type 2 diabetes mellitus (T2DM). Although in some prediabetics (5-10%) plasma glucose levels were found to be elevated and individuals were diagnosed as T2DM, the same amount of people were also recorded as normoglycemic due to a decrease in plasma glucose levels. Klotho is a trans-membrane protein and is being secreted as a soluble protein which can be identified in blood, urine and cerebrospinal fluid. Recent studies have indicated that soluble α-klotho may inhibit the intracellular insulin. Therefore, in our study, we aimed to determine the relationship between serum α-klotho and insulin sensitivity in prediabetics. This study included 26 obese individuals (control group) and 25 obese prediabetics. In order to assess the relation, we determined serum glucose, insulin, homeostatic model assessment of insulin resistance (HOMA-IR) by using autoanalyzer and serum soluble α-klotho levels were measured by using ELISA method. We observed a statistically significant increase in glucose and HOMA-IR levels in obese prediabetes group compared to control group (p<0.05). A significant decrease in insulin and α-klotho were observed in obese prediabetes group compared to control group (p<0.05). The results of correlation analysis revealed that there was a strong and negative significant correlation between glucose and α-klotho (p<0.01). Our results suggest that serum α-klotho may be a valuable marker for the diagnosis of prediabetes. However, further studies are needed to reveal the relationship between prediabetes and α-klotho levels.
