Objective: Prothrombotic and inflammatory status plays an important role in the development of cardiovascular complications of type-2 diabetes mellitus (T2DM) and plants containing biologically active ingredients may be useful for the prevention and management of said complications. The purpose was to investigate the anti-inflammatory, antidiabetic and antiplatelet potential of extracts obtained from fruits of Ribes rubrum L. Materials and Methods: To evaluate the in vitro anti-diabetic and the anti-inflammatory effects of the extracts (ethanol, 50% ethanol and water), their effects on inhibition of α-amylase, α-glucosidase and lipoxygenase enzymes were investigated. In addition, in vitro antioxidant activities and amounts of total phenolic and flavonoid of extracts were examined. For in vivo study, a streptozotocin-induced experimental diabetes model was created in rats. After developed hyperglycemia, the treatment group received ethanol extract of Ribes rubrum (100 mg/kg/day) for 15 days. Plasma glucose levels, tumor necrosis factor-α (TNF-α), platelet P-selectin levels were examined. Results: In the antidiabetic and anti-inflammatory activities assays, the IC50 values of the tested three extracts for α-amylase, α-glucosidase and lipoxygenase were found to be 130 µg/ml, 170 µg/ml, 181 µg/ml; 189 µg/ml, 227µg/ml, 354 µg/ml; 21 µg/ml, 18 µg/ml, 30 µg/ml respectively. Plasma TNF-α (p<0.05) and platelet P-selectin levels (p<0.05) significantly decreased, while plasma glucose level moderately decreased in the treatment group. Conclusion: The results show that fruits of Ribes rubrum may provide support for the prevention/treatment of complications of T2DM by demonstrating its the antithrombotic, anti-inflammatory potential.
